| 摘 要： | 目的: 观察中药有效成分Nordosin对FK506体外抑制致炎细胞因子作用的影响,探讨Nordosin在移植免疫领域存在的潜在应用价值。方法: 建立ConA激活小鼠脾淋巴细胞增殖模型,采用ELISA和RT-PCR方法,测定Nordosin与FK506联合应用对淋巴细胞的IL-2蛋白和mRNA表达的影响;同时,运用流式细胞检测T淋巴细胞CD25+表达的变化。结果: Nordosin与FK506联合用药可以抑制小鼠脾淋巴细胞的IL-2分泌(P<0.01),IL-2mRNA表达以及T淋巴细胞CD25+表达也显著降低(P<0.01),其作用强于单独使用FK506组(P<0.01)。结论: Nordosin可以增强FK506抑制体外淋巴细胞IL-2分泌及其受体表达的作用,Nordosin可望成为一种免疫抑制辅助用药。

| Abstract: | AIM: To investigate the effect of Nordosin in inhibiting proinflammatory cytokine with FK506 in vitro and potential applied value in the transplantation immunity.METHODS: ConA-stimulated T-cell proliferation model was made.Then, the expression of IL-2 both in protein and mRNA lever were investigated by ELISA and RT-PCR.Furthermore, the expression of IL-2 receptor, CD25+ on surface of T lymphocyte was also measured by FCM.RESULTS: Compare with those by FK506 use along, Nordosin enhanced the inhibition of FK506 on the expression of IL-2 significantly (P<0.01).The expression of IL-2 mRNA and its receptor CD25+ were also inhibited by their combining medication (P<0.01).CONCLUSION: Nordosin could enhance the inhibition of FK506 on IL-2 secretion significantly, Nordosin maybe become an immunodepression assistant medication to enlarge the pharmacological effect of FK506. |
