抗肝腹水浸膏对鼠肝纤维化及利尿作用的影响

摘    要:目的:探讨抗肝腹水浸膏对鼠肝纤维化及利尿作用的影响。方法:1、小鼠肝纤维化试验:采用20%、40%CCL4橄榄油溶液,皮下注射制作小鼠肝纤维化模型后给药6周,测定肝组织羟脯氨酸含量,计算肝、脾指数并作病理检查。2、大鼠利尿试验:取试验前禁食18hr大鼠,分别给大鼠灌胃用生理盐水配成的药液50ml/kg,立即放入代谢笼内,收集6hr内的尿液,观察其大鼠6hr的排尿量与灌胃量比值。结果:1、小鼠肝纤维化试验:工艺Ⅱ高、工艺Ⅲ中、Ⅲ高和马洛替酯组小鼠肝组织羟脯氨酸含量下降,工艺Ⅱ中、工艺Ⅲ低和马洛替酯组小鼠血清透明质酸含量下降,工艺Ⅰ高、工艺Ⅱ中和马洛替酯组小鼠脾指数下降,与模型组比较,差异有显著和非常显著性意义(p〈0.05和p〈0.01)。病理组织学检查:工艺Ⅰ低、工艺Ⅱ高、工艺Ⅲ低、Ⅲ中、复方鳖甲软肝片组和马洛替酯片组炎症细胞逐渐减少,点状坏死和变性范围缩小,有明显的纤维组织增生,与模型组比较,差异有显著和非常显著性意义(p〈0.05和p〈0.01)。2、从大鼠利尿试验:工艺Ⅰ低、工艺Ⅱ低、工艺Ⅲ低、呋塞米片组在2hr的排尿量和排尿量与灌胃量比值均增;工艺Ⅰ低、工艺Ⅱ低、呋塞米片组在6hr的排尿量和排尿量与灌胃量比值均增加,但与空白组比较,均无显著性差异(p〉0.05)。结论:工艺Ⅱ供试品,具有抑制肝纤维化及利尿作用。

Abstract:Inorder to explore the effects of hepatic ascites extract on rat hepatic fibrosis and diuresis.Methods:1.mice hepatic test:the model of hepatic fibrosis was established by subcutaneous injection of 20% and 40%CCl4 olive oil solution.The mice was given the extract for 6 weeks.Then,test the content of hyplroxyproline in hepatic tissue,calculate the index of liver and spleen and describe of the liver and spleen pathologic examination.2.rats diuresis test:The rats that had been fasted for 18 hours were given the medicine with salne water(50mg/kg),immediately inmetabolic cage.Collect the urine in 6 hours,calculate the ratio of urine output and intragastric volume.Results:1.mice hepatic test:The content of hydroxyproline in hepatic tissue decreased in processⅡ high,process Ⅲ,process Ⅲ high and marlowe for ester group.The content of serum hyaluronic acid decreased in process Ⅱ,process Ⅲ low and Marlowe for ester group.The index of spleen decreased in process Ⅰ high,process Ⅱ and marlowe for ester group.There is a significant difference compared with model group(p0.05 and p0.01).Pathological organized to examine:In process Ⅰ low,process Ⅱ high,process Ⅲ low,process Ⅲ Fu Fang Bie Jia Ruan Gan Pian group and Marlowe for ester group,the number of inflammatory cells gradually decreased,the range of spotty necrosis and degeneration reduced.There is significant fibrous tissue proliferation.There is significant difference compared with model group(p0.05 and p0.01).2.rats diuresis test:In process Ⅰ low,process Ⅱ low,process Ⅲlow and Furosemide group,the ratio of urine output and intragastric volume increased in 2 hours.In process Ⅰ low,process Ⅱ low and Furosemide group,the ratio of urine output and intragastric volume increased in 6 hours.But there isa non-significant difference(p0.05).Conclusion:the process Ⅱ have the function of inhibiting hepatic fibrosis and diuresis.
