蝉拟青霉代谢产物抑制单胺氧化酶和抗肿瘤活性研究

摘    要:以单胺氧化酶(MAO)为靶标对蝉拟青霉RCEF0947菌株发酵液和菌丝体甲醇乙酸乙酯提取物体外抑制单胺氧化酶活性进行研究,结果发现,RCEF0947 发酵液提取物对MAO-A 具有较强的抑制活性,其对单胺氧化酶MAOA的半数抑制浓度(IC50)为272.97μg/ml。而对于MAO-B 的抑制活性而言,当样品反应浓度为400μg/ml 时,发酵液和菌丝体提取物对MAO-B的IC50 分别为538.37μg/ml 和509.36μg/ml,经统计分析二者之间无显著性差异(p > 0.05)。以中国仓鼠卵巢(CHO)细胞株作为模型,用Resazurin 法检测生长抑制率,对RCEF0947 菌株发酵液和菌丝体提取物进行体外抗肿瘤活性实验,结果发现,两种提取物均具有较强的体外抗肿瘤活性,两种提取物对CHO 细胞株的IC50 分别为122.35μg/ml 和171.93μg/ml。

Abstract:With monoamine oxidase (MAO) as the target, the inhibitories activities of the methanol-ethyl acetate (1:1, V/V) extracts from the broth and mycelia of Paecilomyces cicadae RCEF0947 were investigated. It was found that the broth extract had good inhibitory activity against MAO-A, and the IC50 was 272.97 μg/ml; while for MAO-B, when the reaction concentration of sample was 400 mg/ml, both the extracts from broth and mycelia of RCEF0947 showed strong inhibitory activity with the IC50 of 538.37μg/ml and 509.36 μg/ml (no significant difference between them, p > 0.05), respectively. Meanwhile, a Resazurin cytotoxity assay model of Chinese hamster ovary cells (CHO) was employed to study the anti-tumor activity of extracts from the strain RCEF0947. The results revealed that both the extracts from broth and mycelia of RCEF0947 had strong anti-tumor activity with IC50 of 122.35μg/ml and 171.93μg/ml, respectively.
