正交法优化酸性植酸酶固态发酵工艺

摘    要:以麸皮为基本原料,对黑曲霉TW012 产酸性植酸酶的固态发酵条件进行优化,以提高酸性植酸酶的产量。依据单因素试验和正交试验确定酸性植酸酶固态发酵的最佳工艺条件为:氮源(NH4)2SO4 质量分数1.6%、pH5.0、含水量70%、30℃发酵5d,此时酸性植酸酶的酶活力可达346.576U/g 干基,较优化前提高23.77%。

Abstract:The production process of acidic phytase based on the use of wheat bran as fermentation substrate in solid-state fermentation by Aspergillus niger TW012 was optimized using single factor method and orthogonal array design in this study. The optimum fermentation conditions were found to be: ammonium sulfate content in medium 1.6%, medium initial pH 5.0, medium water content 70%, fermentation temperature 30 ℃, and fermentation cycle 5 days. Under these optimum conditions, the maximum acidic phytase activity in the final fermentation product reached up to 346.576 U/g dry matter, 23.77% higher than before optimization.
