杀线虫海洋枝顶孢霉BH0531发酵条件的响应面优化

摘    要:以自主筛选出的杀线虫海洋枝顶孢霉BH0531为对象,对其发酵条件进行了响应面分析和优化研究。在单因素试验的基础上,采用Plackett-Burman试验筛选出影响杀线虫效果的因素为NaNO3、K2HPO4、发酵时间。通过Box-Behnken设计和响应面分析法优化以上影响因素并对其结果进行分析,最终得到最优发酵条件为NaNO3 3.4 g/L、K2HPO4 1.5 g/L、发酵时间130 h。在此优化条件下,经摇瓶发酵验证,优化后发酵液的杀线活性为88.00%,比优化前提高了22.77%。

Abstract:Using nematocidal marine-derived Acremonium sp. BH0531 screened in the laboratory as research object, the fermentation conditions of the strain were analyzed and optimized by response surface methodology. On the basis of single factor tests, three important factors influencing the nematicidal effect were screened by Plackett-Burman tests as NaNO3, K2HPO4 and fermentation time. The influencing factors and results were analyzed and optimized by Box-Behnken design and response surface analysis method as follows: NaNO3 3.4 g/L, K2HPO4 1.5 g/L and fermentation time 130 h. Under these optimum conditions, through flask-shaking verification tests, it showed the nematicidal activity of the fermentation broth was 88.00%, which was 22.77% higher than that of before optimization.
