摘    要:此次试验建立了高效液相色谱同时定量检测不同大孔树脂苦荞麸皮黄酮富集品中芦丁、烟花苷、槲皮素和山柰酚4种黄酮类化合物含量的方法。分别使用ADS-7、AB-8、D101型大孔树脂富集苦荞麸皮黄酮,采用Intersustain C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈和0.2%磷酸水溶液作为梯度洗脱的流动相,流速1.0 mL/min,柱温35℃,检测波长360 nm。结果显示,芦丁、烟花苷、槲皮素和山柰酚分别在25~400,1.25~20,9.375~150和0.625~10μg/mL的浓度范围内线性关系良好,平均回收率分别为99.48%,100.86%,100.93%和102.27%。结果表明,ADS-7型大孔树脂纯化的苦荞麸皮总黄酮含量较AB-8和D101更高,该方法操作方便、准确性高、重复性好。

Abstract:To establish an assay method for four flavonoids(rutin,kaempferol-3-O-rutinoside,quercetin and kaempferol)in tartary buckwheat enriched by different macroporous resins by using HPLC,three kinds of macroporous resins,including ADS-7,AB-8 and D101 resin were selected as suitable materials to purify flavonoids from tartary buckwheat.Intersustain C18 column(4.6 mm×250 mm,5μm)was used with mobile phase consisting of acetonitrile and 0.2%phosphoric acid at a flow rate of 1.0 mL/min.The detection wavelength was set at 360 nm,and the column temperature was 35℃.The results showed that rutin,kaempferol-3-O-rutinoside,quercetin and kaempferol all had good linear relationships in the range of 25-400,1.25-20,9.375-150 and 0.625-10μg/mL,and the average recoveries were 99.48%,100.86%,100.93%and 102.27%,respectively.The results also showed that the content of flavonoids in tartary buckwheat enriched by ADS-7 macroporous resin was higher than that of AB-8 and D101 macroporous resins.In conclusion,the method used to determine four enriched flavonoids in tartary buckwheat was simple,accurate and reproducible.
