纤维素酶高产菌筛选鉴定及酶学性质初步研究

摘    要:该研究从土壤中筛选出一株纤维素酶高产菌株SP08,并通过菌落形态特征和ITS测序技术对菌株进行鉴定,最后通过酶促反应初步测定了该菌所产纤维素酶的性质。结果表明,筛选得到菌株SP08,经鉴定为康氏木霉(Trichoderma koningii),该菌株具有较高的纤维素酶产酶能力,培养60 h时纤维素酶活性即达(18.54±0.75),72 h时酶活达到最高(19.18±0.68)U/mL。酶学研究表明,菌株SP08所产纤维素酶最适温度和pH分别为50 ℃和5.5,在20~50 ℃及pH4.0~6.0时稳定性较高;Mg2+、Mn2+和Ca2+能够提高该酶活性,而Fe2+和Cu2+却对该酶具有抑制作用。

Abstract:A high cellulase-producing strain SP08 was screened from soil, and identified based on the morphology characteristics of colony and the ITS sequencing technology. Finally, the properties of cellulase produced by the strain were preliminarily determined by enzymatic reaction. The results showed that strain SP08 was screened out and identified as Trichoderma koningii, which had high cellulase production ability. The cellulase activity reached (18.54±0.75) U/ml when cultured for 60 h, and reached the highest (19.18±0.68) U/ml for 72 h. The results of enzymology studies showed that optimum temperature and pH of cellulase produced by strain SP08 were 50 ℃ and 5.5, respectively, with high stability at 20-50 ℃ and pH 4.0-6.0. Mg2+, Mn2+ and Ca2+ could increase cellulase activity, while Fe2+ and Cu2+ had inhibitory effects on cellulase.
