动物双歧杆菌乳亚种菌株HCS04-002益生特性的研究

摘    要:该研究测定动物双歧杆菌乳亚种(Bifidobacterium animails subsp. lactis)菌株HCS04-002对酸、胆盐、人工胃液和人工肠液的耐受性,并采用邻硝基苯-β-D-吡喃半乳糖苷(ONPG)法及甘油三酯试剂盒分别测定菌株的β-半乳糖苷酶活力和甘油三酯降解能力。结果表明,菌株HCS04-002在pH 2.0和pH 3.0环境下处理17 h,存活率达60%以上;在0.3%、0.5%、1.0%、1.5%的胆盐环境处理17 h,存活率达70%以上;经人工胃液和人工肠液消化后,存活率达99%以上;β-半乳糖苷酶活力为0.78 U/mL;甘油三酯降解率为43%。综上,动物双歧杆菌乳亚种菌株HCS04-002具有较好的耐酸、耐胆盐及耐胃肠液能力,进入人体后能够以较高存活率到达肠道发挥作用,并具有一定的缓解乳糖不耐受和降甘油三酯功能。

Abstract:In this study, the tolerance of Bifidobacterium animails subsp. lactis HCS04-002 to acid, bile salts, artificial gastric fluid and artificial intestinal fluid was measured, and the β-galactosidase activity and triglyceride degradation ability of the strain were determined by O-nitrophenyl-β-D-galacto-pyranoside (ONPG) method and triglyceride kit, respectively. The results showed that the survival rate of strain HCS04-002 was over 60% after the strain was treated at pH 2.0 and pH 3.0 for 17 h. The survival rate of strain HCS04-002 was over 70% after the strain was treated for 17 h in 0.3%, 0.5%, 1.0%, and 1.5% bile salt environment, respectively. After digestion with artificial gastric fluid and artificial intestinal fluid, the survival rate was over 99%. The activity of β-galactosidase was 0.78 U/ml, and the triglyceride degradation rate was 43%. In summary, B. animails subsp. lactis HCS04-002 showed good acid, bile salt, and gastrointestinal fluid tolerance. After entering the human body, the strain could reach the intestine with a higher survival rate and play a role, and had a certain alleviating action on lactose intolerance and triglyceride-lowering function.
