白芍总苷对高尿酸血症肾损害大鼠肾脏的保护作用

摘    要:目的:探讨白芍总苷(total glucosides of paeony, TGP)对高尿酸血症肾损害(hyperuricemic nephropathy, HN)大鼠肾脏的保护作用。方法:雄性SD大鼠50只,随机分为5组:正常组、模型组、TGP低、高剂量组(100、300 mg·kg?1·bw?1)、别嘌醇组(27.0 mg·kg?1·bw?1),每组10只。每天上午7:00和晚上20:00,模型组、TGP低、高剂量组和别嘌醇组分别灌胃腺嘌呤100 mg/kg+氧嗪酸钾1500 mg/kg混悬液制造HN模型,每天中午12:00按剂量灌胃给药TGP和别嘌醇,正常组和模型组灌胃给予等量生理盐水,持续5周。每隔1周检测各组大鼠体重、血清尿酸、肌酐、尿素氮等指标,5周后,检测大鼠双肾重与体重的比值,同时对肾进行病理切片观察。结果:第5周时,与模型组相比,TGP高剂量组能显著降低HN模型的肾重(P<0.01)、肾体比(P<0.01),降低HN模型血清中的尿酸(P<0.01)、尿素氮(P<0.05)和肌酐(P<0.01)水平,缓解HN模型肾小球系膜细胞增生,缓解肾小管扩张,减少肾小管间质损伤和肾脏炎性细胞浸润。结论:TGP对HN大鼠的肾脏有一定的保护作用。

Abstract:Objective: To investigate the renal-protective functions of total glucosides of paeony(TGP) on hyperuricemic renal damage(HN) rats. Methods: Fifty male SD rats were randomly divided into 5 groups: Normal group, model group, TGP low and high dose groups(100 and 300 mg·kg?1·bw?1), and allopurinol group(27.0 mg·kg?1·bw?1), with 10 rats in each group. At 7:00 a.m. and 20:00 p.m. every day, the model group, TGP low-dose group, TGP high-dose group and allopurinol group were given adenine 100 mg/kg+oteracil potassium 1500 mg/kg suspension respectively to make HN model. TGP and allopurinol were intragastrically administered at 12:00, the normal group and model group were given the same amount of normal saline every day. The observation time was 5 weeks. The body weight, serum uric acid, creatinine, urea nitrogen and other indicators of rats in each group were detected every two weeks. After 5 weeks, the ratio of bilateral kidney weight to body weight and the pathological section of kidney were observed. Results: At the 5th week, compared with the model group, TGP high-dose group could significantly reduce the renal weight(P<0.01), renal body ratio(P<0.01), serum uric acid(P<0.01), urea nitrogen(P<0.05) and creatinine(P<0.01) levels of HN model, alleviate the proliferation of glomerular mesangial cells, alleviate the expansion of renal tubules, and reduce tubulointerstitial injury and renal inflammatory cell infiltration of HN model. Conclusion: TGP had a protective effect on the kidney of HN rats.
