粟酒裂殖酵母Atp25蛋白定位和功能研究

通过荧光显微镜观察、线粒体提取、基因敲除和免疫印迹等技术,研究粟酒裂殖酵母Atp25蛋白定位和Atp25蛋白缺失对线粒体功能的影响. Atp25是核基因组编码蛋白质,生物信息学预测和荧光显微镜观察结果显示Atp25蛋白定位在线粒体中,免疫印迹技术进一步验证了Atp25蛋白的定位. 表型研究结果显示,Δatp25菌株存在生长及呼吸缺陷. Δatp25菌株中线粒体呼吸链复合体亚基蛋白(Cox1、Cox2、Cox3、Cob1、Atp6)水平显著降低. 实时荧光定量PCR结果表明,Δatp25菌株中这些复合体亚基的mRNA水平没有变化.

Study on the Localization and Function of Atp25 Proteinin Schizosaccharomyces Pombe

In this paper,we investigate the localization of Atp25 protein in Schizosaccharomyces pombe and the effect of Atp25 protein deletion on mitochondrial function by fluorescence microscopy,mitochondrial extraction,gene knockout and immunoblotting. Atp25 is a nuclear genome-encoded protein. Bioinformatics predictions and fluorescence microscopy show that Atp25 protein is localized in mitochondria. The localization of Atp25 protein is further verified by immunoblotting. Phenotypic studies reveal growth and respiratory defects in strain Δatp25. The levels of mitochondrial respiratory chain complex subunit proteins(Cox1,Cox2,Cox3,Cob1,Atp6)are significantly reduced in strain Δatp25. Real-time fluorescence quantitative PCR results show no change in the mRNA levels of these complex subunits in strain Δatp25.
