一株产菊粉内切酶酵母Kluveromyces NSY5发酵条件优化

摘    要:在单因素实验基础上,采用Plackett-Burman实验设计对影响KluveromycesNSY5发酵产菊粉内切酶的碳源、氮源、无机盐、发酵时间等8个选择因素进行筛选,结果显示,培养基的菊芋汁浓度、摇瓶装液量和发酵时间对菌株发酵产酶具有显著影响。根据实验结果和经验方法对显著影响因素的优化取值范围进行预估,并选择适当的水平设计,用Box-Behnken中心组合实验和响应面寻优分析找出显著影响因素的极优值,验证实验结果表明,采用优化的菊芋汁浓度3.5%、装液量50 mL/250 mL、发酵时间56 h,KluveromycesNSY5摇瓶发酵液中的菊粉内切酶I/S比值达到18.29,比初始发酵条件下的菊粉内切酶I/S比值12.4提高47.5%,优化预测值和实验验证值的相对误差为0.26%。
