吸附树脂AB-8精制栀子黄色素的工艺研究

摘    要:采用大孔吸附树脂柱层析柱分离天然产物栀子黄,以AB-8大孔吸附树脂为研究对象,探讨了树脂对栀子黄的静态吸附率和吸附流速、洗脱流速、流脱方式对树脂吸附的影响,得到AB-8树脂吸附栀子黄较为合适的工艺.以AB-8树脂为吸附剂,采用超声波提取法获得的色素粗提液为吸附液,吸附流速1.5mL/min,并采用梯度洗脱法对柱上色素进行了洗脱,洗脱流速2.5mL/min.精制后色素溶液A238/A440从2.12降到0.64,精制色素得率89.27%,色素色价达到221.24.

Abstract:The separation result of the crocin with macroporous absorption resins column chromatography was studied.Regarding AB-8 macroporous adsorption resin as the research object,the experiments on static adsorption rate of the resin for crocin,the flow rate on absorption,the flow rate on desorption and the solvent of desorption on the behavior of the resin adsorption were made and the optimum parameters were obtained as follows:AB-8 resin as absorbent, the gardenia yellow pigment with ultrasonic extraction as elution program, 1.5ml/min as the flow rate on absorption, 2.5ml/min as the flow rate on desorption. The A_(238)/A_(440) of the resulted product was lowered to (0.64) from 2.12, and the yield and the color value of the extracted pigment achieved 89.27% and 221.24 respectively.
