酸果蔓中SOD酶的提取及活性测定

摘    要:利用正交法探讨了磷酸缓冲液用量、pH值、提取时间、丙酮用量对酸果蔓SOD酶提取率的影响,并用改良Marklund法测定了酸果蔓中SOD的活性,当在酸果蔓中加入1.5倍的磷酸缓冲液,pH7.8,提取2h,再用1倍体积的丙酮沉淀,酸果蔓中SOD酶粗品的提取率达0.95%;酸果蔓中SOD酶单位体积的活力为433.7U/mL。酸果蔓中SOD酶含量高且活性强。

Abstract:Effect of extraction rate on Cranberry superoxide dismutase (SOD) were discussed using orthogonal design, in which four factors have considered,that is,amount of phosphoric acid buffer solution(PBS), PH, extraction time and amount of acetone. activity of Cranberry SOD was determined by Marklund method. Experimental results were as fdlows:Extraction rate of Cranberry SOD up to 0.95%, when add 1.5 time of phosphoric acid buffer solutions in Cranberry, pH7.8, extraction 2 h and 1 time of volumes the acetone precipitations. Cranberry SOD unit volume vigor is 433.7 U/mL. Cranberry has a very high SOD content with high activity.
