摘    要:以有机膦鞣革屑为原料,采用碱-酶两步法提取胶原蛋白,单因素实验结果表明:在m(氧化镁)∶m(革屑)=0.06∶1,反应温度70℃,反应时间2.5 h,m(碱性蛋白酶)∶m(革屑)=0.006∶1的条件下,提胶率达88%。采用多媒体显微镜、红外光谱仪以及凝胶渗透色谱仪对最优条件下提取的胶原蛋白进行了结构分析,并与市售的明胶水解产物做了分析对比。结果表明:提取的胶原蛋白与市售的明胶水解产物的谱图基本一致,市售的明胶水解产物的平均相对分子质量为1 346和564,而提取的胶原蛋白的平均相对分子质量较大,为1 928和755,说明提取的物质为胶原多肽和氨基酸的混合物。应用实验结果表明:当胶原多肽溶液的用量为80 mL/m3时,甲醛去除率可达81%。

Abstract:The two-step method with alkali and alkaline protease is used to hydrolyze phosphine tanned leather shavings.The best hydrolyzation conditions are:m(magnesium oxide)∶m(leather shavings)=0.06∶1,reaction temperature 70 ℃,reaction time 2.5 h,and m(alkaline protease)∶m(leather shavings)=0.006∶1.Under these conditions the extraction rate of collagen reaches 88%.Multimedia microscope,FTIR and GPC are used to analyze the extracted collagen and the commercial glutin hydrolysate.The results indicate that the FTIR spectra of collagen and glutin hydrolysate are nearly the same.The average relative molecular weights of glutin hydrolysate are 1 346 and 564,but the average relative molecular weights of extracted collagen are 1 928 and 755,bigger than those of glutin hydrolysate.These all show that the extracted substance from leather shavings is the mixture of collagen peptide and amino acid.Moreover,the application result indicates that when the dosage of collagen peptide is 80 mL/m3,formaldehyde removal rate is 81%.
