纤维联接蛋白FN、凝血酶敏感蛋白(TSP)在成纤维细胞及骨髓基质细胞的免疫电镜定位

摘    要:本文用免疫电镜技术(过氧化酶标记和胶体金标记)在来自兔角膜瘢痕的纤维母细胞和来自小鼠骨髓的长期培养的基质上进行了纤维粘连蛋白(FN),血小板凝血酶敏感蛋白(TSP)定位。结果显示在纤维母细胞和骨髓基质细胞表面以及细胞外细纤维构成的网中。另外,有时在上述二种细胞胞浆中也可以看到FN和TSP的定位。应用免疫电镜技术可以研究各种结缔组织增生性疾病,FN和TSP的分布以及它们和结缔组织增生的关系,为纤维化形成机制以及胶原形成细胞、基质细胞和其它细胞的关系提供了形态学依据。

Abstract:Resort to immunoelectron microspic technique(peroxidase labelling and colloidal gold maker),we can sure- ly localize fibronectin(Fn)and thrombospondin(TSP)on cultured fibroblasts derived from rabbit corneal scars and on mouse bone marraow stromal cells. The studies reported here showed that the fibroblasts and stromal cells factually synthesized and secreted Fn and TSP which were found to appear on the cell surface and interspace among the adjacent cells as well as the extracellular fibrous network matrix,demonstrating some bulky patch patterns.In addition,both the fibroblasts of rabbit corneal scars and the stromal cells of mouse bone marrow in long-term culture contain fibronectin and thrombospondin in their cytoplasms. Application of immunoelectron microscopic technique makes it possible to study the localization and distri- bution of Fn and TSP in the varied diseases concerning the connective tissue hyperplasia.It could provide a direct means to present visual morphological evidence about the mechanism of fibrosis formation and the interaction of collagen-forming cells,stromal cells,and other type cells.
