荧光RT-PCR与常规RT-PCR检测禽流感病毒比较研究

摘    要:采用目前公认的2种敏感特异的检测方法———荧光RT-PCR和常规RT-PCR———检测标准毒株禽流感病毒H9亚型(鸡胚尿囊液)及部分待检样品,进行灵敏度、特异性、安全性等方面的比较.结果表明:荧光RT-PCR方法在4 h内就能得出结果,检测尿囊液的禽流感病毒灵敏性达到10-7;RT-PCR方法6 h出结果,其灵敏度为10-5,且荧光RT-PCR不需电泳,不接触EB,更安全.因此,荧光RT-PCR比常规RT-PCR更灵敏、快速和安全.

Abstract:An experiment with the allantoic fluid of AIV H9 subtype and chicken samples is conducted to compare two accepted sensitive methods of fluorescence RT-PCR and RT-PCR on sensitivity,specificity and reliability.Fluorescence RT-PCR AIV could be detected in four hours with a dilution of 10~(-7) allantoic fluid of AIV H9 subtype;RT-PCR test could detect with only a dilution of 10~(-5) allantoic fluid of AIV H9 subtype in six hours.Fluorescence RT-PCR has an advantage of omitting the step of electrophoresis,which makes it safer than RT-PCR without getting touch with EB.Here a conclusion can be safely drawn that fluorescence RT-PCR appears to be more sensitive,faster and safer than the RT-PCR methods for detection of AIV.
