应用减毒株和强毒株试制脊髓灰质炎灭活疫苗的比较

摘    要:作者采用单层猴肾细胞,接种中Ⅲ_2 及 Leon 株毒种。收获后的病毒悬液经澄清过滤,超滤浓缩后用 Sepharose CL 6B 凝胶过滤,再用 DEAE-Sephadex A_(50)进行离子交换层析提纯。最后加1/4000福尔马林,放37℃水浴灭活13天,制成灭活疫苗。疫苗中残余牛血清均低于60ng/ml,D-Ag 含量中Ⅲ_2 为2316Du/ml,回收率为64%;Leon 株为2068Du/ml,回收率为71%。分别免疫家兔均产生相当高(1024~8192)的血清中和抗体。

Abstract:Monolayer cells of Rhesus Monkey kidney were inoculated with attenuated Zhong Ⅲ_2and wild Leon strains separately.When CPE Completely occured,the virus suspensions were harvest-ed.The virus suspension was clarified by a series of membrane filtration.Finally it was filtered witha membrane of 0.2μm.After clarification,the virus suspension was concentrated about 300 to 500folds by ultrafiltration.The concentrated virus suspension was purified by chromatography withSepharose CL 6B column and by ion-exchange chromatography with DEAE-Sephadex A-50 column.The purified virus suspension was inactivated with 1:4000 formalin at 37℃ for 13 days.The deter-ruination of D-Antigen was carried out by means of neutralizing antibody combining test (NAST).Theresults indicated that the attenuated and wild strain IPV contain 2316 D-Antigen Units (DU)/ml re-spectively.The residual calf serum protein of IPV was assayed by means of radioimmunoassay.Theamount of calf serum protein in all purified virus samples was less than 60 ng/ml.Two kinds of IPVwere used for immunizing rabbits.High titers of specific neutralizing antibody were obtained (rangedfrom 1024 to 8192).The results reported in this paper indicated that the similar results have been obtained for each stepof preparation of IPV with two strains.Also it gave some basic information which oould be useful infurther study on maidng IPV with attenuated poliovirus strains.
