抗大肠杆菌荧光抗体的制备及其特性

摘    要:活体大肠杆菌与弗氏不完全佐剂乳化后注射到新西兰大白兔体内15d后,利用两步沉淀法提取并纯化得到大肠杆菌表面蛋白抗体IgG,与FITC的交联率F/P达到2.7,未发现FITC对大肠杆菌表面蛋白抗体的特异性有显著的影响。将抗大肠杆菌荧光抗体注入小白鼠体内后,除肠道内有荧光显示外,肝脏、心脏、肾脏中未发现荧光显示,表明大肠杆菌只在小白鼠大肠存在,根据观测到的荧光强度可定性和定量地判断大肠中的大肠杆菌。

Abstract:E.coli in vitro with incomplete Freund′s adjuvant was emulsificated and injected into New Zealand white rabbits.Using two-step methods of precipitation,IgG of anti-protein of E.coli cell surface was successfully extracted and purified from the rabbits in 15 d after injection.The rate of the IgG cross-linked with FITC reached to 2.7.It was not found significant effect on the specificity of the antibody to E.coli surface protein.When the FITC-antibody of anti-E.coli was injected into mice,fluorescence was observed only in the large intestine rather than other organs or tissues such as the liver,heart,and kidney.The results showed that E.coli existed only in the mouse large intestine and the FITC-antibody of anti-E.coli could be used to examine and determine E.coli qualitatively and quantitatively.
