响应面法优化无花果中蛋白的提取条件

摘    要:采用单因素试验方法,研究磷酸盐缓冲液浓度、料液pH、固液比对无花果中蛋白提取量的影响,并利用Design-Expert软件响应面分析法构造模型,确定无花果中蛋白的最优提取条件为:磷酸盐缓冲液浓度0.08 mol/L,pH 7,固液比1∶50,无花果蛋白提取量的最优预测值为9.633 27 mg/g,试验验证,所得蛋白提取量的实际值为9.475 0 mg/g。

Abstract:To optimize the extraction condition of protein from ficus carica,the effects of phosphate concentration,pH,ratio of solid and liquid phase were investigated by single-factor experiment.Response surface methodology from Design-Expert software was employed to formulate a model to optimize extraction condition.The results showed that the phosphate concentration was 0.08 mol/L,the pH was 7,the ratio of solid and the liquid phase was 1∶50,the optimization value could be obtained and the predicted value was 9.633 27 mg/g,under the same condition,the actual extraction quantity of protein was 9.475 0 mg/g.
