玉米皮水解液生产单细胞蛋白与L-阿拉伯糖

摘    要:利用酵母菌发酵玉米皮稀酸水解液中的葡萄糖和木糖生产单细胞蛋白(SCP),再从发酵上清液中分离制备L-阿拉伯糖。采用正交实验方法确定了玉米皮稀硫酸水解的最佳工艺条件:H2SO4质量分数为1.5%,水解温度120℃,水解时间3 h,固液比1:10(g:mL)。在此条件下,水解得到木糖、阿拉伯糖和葡萄糖含量分别为22.17 g/L、12.29 g/L、11.16 g/L。比较了四种酵母菌对木糖和阿拉伯糖的利用情况,结果表明,热带假丝酵母利用木糖速度最快,菌体生物量最高,且发酵过程中阿拉伯糖损失较小。利用该菌发酵玉米皮酸水解液,菌体生物量达12.6 g/L,L-阿拉伯糖晶体得率为6.8%(以玉米皮干重计)。

Abstract:The single-cell protein(SCP)can be produced by using yeast to ferment glucose and xylose which were from the corn husk hydrolyzate.The crystalline L-arabinose was obtained from the fermentation supernatant.In the orthogonal experiment of sulfuric acid hydrolysis,the optimal conditions were as follows: sulfuric acid mass fraction 1.5 %,hydrolysis temperature 120℃,hydrolysis time 3 h,the ratio of solid to liquid(W/V) was 1∶10.Under such conditions,the yield of xylose,arabinose and glucose was 22.17 g/L,12.29 g/L,11.16 g/L,respectively.The fermentation performance of xylose and arabinose of four kinds of yeast was examined.The results show that Candida tropicalis had the highest metabolism rates and yield of biomass,in addition,the loss of the L-arabinose is relatively low.The yield of biomass and L-arabinose was 12.6 g/L and 6.8 %,respectively.
