摘    要:基于原花青素与香草醛的显色反应 ,采用分光光度法对葡萄籽提取物中原花青素含量的测定进行了研究 ,对标准物的选择、催化剂添加量和反应时间进行了讨论 ,确定了香草醛 盐酸法的原花青素测定方法。该方法检测波长为 5 0 0nm ,反应时间为 15h ,反应温度为(2 0± 1)℃ ,以儿茶素为标样 ,线性范围为 10~ 15 0 μg/mL ,相对标准偏差为 0 78%~ 0 79% ,加标回收率为 97 4%~ 98 3 %

Abstract:Based on the color developed reaction of proanthocyanidin with vanillin, spectrophotometric method was used to analysis the proanthocyanidin from grape seed extracts. The choices of standard samples, catalyst amount and the reaction time were discussed, and the vanillin hydrochloric acid method was developed. As a result, catechin was used as the standard sample for the determination of proanthocyanidin. Under the conditions of 500 nm wavelength, (20±1)℃ and 15 h, the linearity range was 10~150?μg/mL, and the relative standard deviation and recovery was in the range of 0.78%~0.79% and 97.4%~98.1% respectively.
