氟伐他汀对高脂血症兔心肌缺血再灌注损伤心肌局部细胞间粘附分子-1的影响

摘    要:目的: 研究氟伐他汀对高脂血症兔心肌缺血再灌注损伤心肌局部细胞间粘附分子-l(ICAM-l)表达的影响。方法: 30只高脂喂养的日本大耳白兔随机分为3组(n=10):缺血再灌注组;氟伐他汀组,缺血再灌注前3 h,氟伐他汀10 mg·kg-1 口服给药;假手术组。实验中监测心电图及心功能,实验完毕取心肌组织,Evans蓝及氯化三苯基四氮唑(TTC)染色测心肌梗死程度,逆转录聚合酶链反应法(RT-PCR)测心肌局部ICAM-1 mRNA表达。结果: 缺血再灌注后,与缺血再灌注组相比,氟伐他汀组心肌局部ICAM-ImRNA表达降低(P <0.01),心肌梗死程度降低(P <0.01),心功能显著改善。结论: ICAM-1mRNA表达增高是心肌缺血再灌注损伤的重要因素,氟伐他汀短期预治疗可降低ICAM-1 mRNA表达,降低心肌梗死程度,改善心功能。

Abstract:AIM: To investigate the effects of fluvas-tatin on expression of intercellular adhesion molecule-1 af-ter myocardial ischemia-reperfusion in rabbits with hyper-lipidemia. METHODS: 30 rabbits which were gastric perfusion administered intralipid were randomly divided into 3 groups (n=10 in each); IR (ischemia-repeifu-sion) group, S (sham-operation) group and F (fluvastatin 10 mg·kg-1) group. Electrocardiography and cardiac function were recorded during the experiment. At the end of reperfusion, ischemic area and infarct size were defined by Evans blue and triphenyltetrazolium chloride staining. The expression of ICAM-1 in myocardium was measured by RT-PCR. RESULTS: After ischemia-reperfusion, the expression of ICAM-1 mRNA in myocardial and infarct size decreased and cardiac function significantly improved in F group compared with IR group. CONCLUSION: The increase of expression of ICAM-1 mRNA in myocardi-al may be one of the important factors in inducing myocar-dial ischemia-reperfusion injury. The myocardial protec-tive mechanism of fluvastatin maybe attribute to its effect on decreasing the expression of ICAM-1 mRNA in myo-cardial.
