麦冬多糖OPF-1的结构分析

摘    要:分离纯化麦冬多糖,得到多糖组分OPF-1,并对其一级结构进行初步分析。水提醇沉法得到麦冬粗多糖,Sevage法除蛋白后经Sephadex G-100柱分离纯化得到OPF-1。运用完全酸水解和HPLC法测定麦冬多糖OPF-1的单糖组成;并采用高碘酸氧化、Smith降解、GC、IR等方法研究OPF-1的化学结构。结果表明,麦冬多糖OPF-1为果聚糖,其相对分子质量为48000,主要由2→1和2→6糖苷键连接的呋喃型果糖组成。该多糖中还含有少量的葡萄糖(果糖与葡萄糖的摩尔比为16∶1),可能连接在多糖的还原性末端。

Abstract:The structure of a polysaccharide OPF-1 obtained from the Ophiopogon japonicus polysaccharide was analized.OPF-1 was obtained using sephadex G-100 column,following water extract of Ophiopogon japonicus was precipitated by ethanol and deproteinized by method of Sevage.The chemical structure of OPF-1 was analized using methods of complete hydrolysis,HPLC,periodate oxidation,Smith degradation,GC and IR et al.The results indicate that the relative molecular mass of OPF-1 was estimated to be 48 000,which was mainly composed of Fruf by linkage of 2→1 or 2→6.The polysaccharide OPF-1 contains trace glucoses [n(fructose)∶n(glucose)=16∶1],which maybe connected to its reducing teminal.
