响应面法同步优化裙带菜中蛋白和多糖提取工艺

摘    要:在单因素试验基础上,以裙带菜蛋白和多糖得率为响应值,利用响应面法对裙带菜中蛋白和多糖提取工艺进行同步优化。研究表明,碱液质量分数、提取时间、提取温度和液料比4个因素对裙带菜蛋白和多糖得率均有显著影响。根据响应面法,绘制三维及等高线叠加图,应用同步优化程序,得出裙带菜蛋白和多糖得率均高的最佳提取工艺参数为碱液质量分数2.2%、提取时间5h、提取温度55℃、液料比23:1(mL/g)。在此条件下进行验证实验,得到裙带菜蛋白得率为63.21%,裙带菜多糖得率为29.11%,与数学模型计算所得预测值接近。可见,响应面同步优化法对裙带菜蛋白和多糖提取条件进行同时优化合理可行。

Abstract:Response surface methodology was used to optimize the simultaneous alkaline extraction of proteins and polysaccharides from Undaria pinnatifi da based on extraction effi ciency. All four operating parameters including alkaline concentration, extraction time, extraction temperature and solvent-to-solid ratio on had a signifi cant effect on the extraction yields of proteins and polysaccharides. The optimum process conditions for simultaneously increasing the yields of proteins and polysaccharides were found to be extraction at 55 ℃ for 5 h with an alkaline concentration of 2.2% and a solvent-tosolid ratio of 23:1 (mL/g). The actual yields of proteins and polysaccharides under the optimized conditions were 63.21% and 29.11%, respectively. Good agreement with the predicted values was found. Therefore, response surface methodology is a feasible way to optimize the simultaneous alkaline extraction of proteins and polysaccharides from Undaria pinnatifi da.
