摘    要:冠心病是一种由遗传和环境共同作用引起的复杂性疾病。大量研究表明细胞色素P450(CYP)代谢物如环氧二十碳三烯酸(EETs)、20-羟基二十碳四烯酸(20-HETE)和性激素在维持心血管功能稳态中起重要作用。在体内,CYP2C19和CYP2J2可催化花生四烯酸代谢生成EETs;CYP4F2和CYP1A1是20-HETE生成的主要合成酶; CYP17A1和CYP19是性激素合成的重要酶。因此,上述CYP基因的多态性很有可能通过影响其代谢产物的生成进而影响冠心病的发生。本文对近5年CYP基因多态性与冠心病易感性关联研究作一综述,以期为冠心病个体化防治提供参考。

Abstract:Coronary heart disease (CHD) is caused by genetic environmental interaction. Studies have shown that cytochrome P450 (CYP) metabolites such as epoxyeicosatrienoic acids (EETs), 20-hydroxy-eicosatetraenoic acid (20-HETE) and sex hormones play important roles in maintaining cardiovascular homeostasis. In vivo, CYP2C19 and CYP2J2 catalyze the metabolism of arachidonic acid (AA) to EETs; CYP4F2 and CYP1A1 are the main synthetases of 20-HETE; CYP17A1 and CYP19 are important synthetases of sex hormones. Therefore, polymorphism of the above CYP genes is likely to affect the development of CHD by influencing the production of its metabolites. This article reviews the five years' advances in the relationship between cytochrome P450 SNP and susceptibility to coronary heart disease so as to provide referential ideas for the personalized treatment of CHD.
