摘    要:以鸡血红细胞悬液为研究模型,通过在对其进行慢速冻结的过程中引入交变电场,研究了交变电场对鸡血红细胞冻结特性的影响。结果表明在1 MHz交变电场作用下,经历了冷冻速率为5℃/min的慢速冷冻后部分红细胞没有出现细胞膜破裂现象,细胞保持了完整的形态,细胞存活率为23.4%±1.43%,比对照组明显提高;但同使用质量分数为3.3%的低温保护剂(二甲基亚砜)的保存效果(37.2%±2.81%)相比还较差。可见将交变电场应用于生物材料低温保存是一种新颖有效的低温保存技术,但还是需要进一步研究最佳电场条件以提高低温保存效率。

Abstract:Chicken red blood cell suspension was selected as research subject,the effect of alternating electric field introduced during process of slow freezing on its freezing characteristic was studied.The results show that the cell membrane rupture phenomenon didn’t appear in some red blood cells after cryo-preservation at 1 MHz alternating electric field,these cells maintain intact configuration and the final cell viability was 23.4%±1.43%,which is significantly higher than the control group.However,compared with the group using low-temperature protective agent of DMSO for ass fraction of 3.3%,the result was still poor.It indicates that alternating electric field is a novel and effective technology in preserving biological materials,but needs further study to find the optimal condition to improve cryo-preservation efficiency.
