茶叶多糖的提取纯化及其单糖组分的鉴定

摘    要:研究了用水煮醇沉法提取茶叶多糖,用savage法除蛋白,苯酚-硫酸法测多糖含量,通过正交试验确定茶叶多糖的最佳提取条件;粗多糖经Sephadex-100柱层析纯化,得到精制茶叶多糖;薄层层析法鉴定单糖组分。结果表明:最佳提取条件为95℃,料液比1∶20(g:mL),提取时间2h,提取3次,茶叶多糖含量为35.92mg/g。茶叶多糖中含有2种不同的多糖,水解得到单糖D-半乳糖、D-甘露糖、D-葡萄糖、L-阿拉伯糖等。

Abstract:A water-soluble polysaccharide was extracted from Tea Leaves by of boiling water then ethanol deposition. deproteinized by Seveg and determined the content of polysaccharides with phenol-sulfuric acid. The optimal condition was determined by the orthogonal experiment. The crude polysaccharides were isolated and purified by chromatographying with Sephadex G-100 column, The results showed that the optimal extraction conditions were 95℃, 2 h, 3 times extraction as well as the ratio 1∶20 (w/v). Under this condition 35.92 mg/g(DW) polysaccharides was obtained. The Tea polysaccharides had two different polysaccharides which contain D-galactose, D-mannose, D-glucose and L-arabinose after hydrolysis.
