聚乙二醇化重组人红细胞生成素在动物体内的作用

摘    要:目的 研究聚乙二醇化重组人红细胞生成素(PEG-rhEPO)在动物体内的作用。方法 经动物皮下注射后比较PEG-rhEPO与普通rhEPO在网织红细胞百分数(RET%)、用药频度、肾性贫血纠正以及对病模动物的保护作用。结果 PEG-rhEPO组网织红细胞百分数升高持续时间可达7d以上,而rhEPO组只能达到4d;在周剂量(0.08μg/只)相同的条件下,PEG-rhEPO每周给药1次小鼠红细胞比容(HCT)增高15.3%,与rhEPO每周给药3次的HCT增高13.4%效果相当,而与rhEPO每周用药1次的HCT增高6.0%有明显差异。在病模大鼠的贫血治疗实验中,PEG-rhEPO不仅每周1次给药能达到rhEPO每周3次给药的效果,而且对患病大鼠的死亡保护作用也远优于rhEPO。结论 PEG-rhEPO作用时间持久,用药次数少,纠正贫血效果明显,保护效果良好。

Abstract:Objective To study the effect of PEG-rhEPO in animals.Methods Inject s.c. PEG-rhEPO into animals and compare the percentage of reticulocyte( RET% ) .frequency of administration .rectification of renal anemia as well as the protection against death with those injected with rhEPO. Results The increase of RET% of the animals injected with PEG-rhEPO lasted for more than 7 days,but that with rhEPO for only 4 days. At the same dosages(0.08ug/mouse/week) , the hematocrit(HCT) of mice injected with PEG-rhEPO once a week increased by 15. 3% ? which was not significantly different from that with rhEPO 3 times a week(13.4%) .however,it was significantly higher than that with rhEPO once at week(6.0%) .In rat model of renal anemia, the curative effect of PEG-rhEPO injected once a week was equal to that of rhEPO injected 3 times a week, and the protective rate of the former was also significantly higher than that of the latter. Conclusion Compared with rhEPO, PEG-rhEPO showed long action time, low administration frequency, significant rectificating effect on anemia and good protection agaist death.
