恒温PCR双通道荧光读数仪的系统设计

摘    要:针对传统PCR检测仪在检测速度和精度上存在的问题,文中设计了恒温PCR双通道荧光读数仪系统,系统采用实时荧光核酸恒温扩增检测技术(SAT),大幅提高了扩增反应时间。然而考虑到SAT技术仍无法避免假阴性,文中在此基础上提出了双通道的光学系统设计,在检测样品阴阳性的同时对阴性样品进行实时监控。结果表明,该系统最快可在30 min内完成检测,可有效指示假阴性现象的出现,并具有检测速度快、精确度高、且稳定性好等优点。

Abstract:A system of constant temperature dual-channel fluorescent reader is designed to improve the detection speed and accuracy of traditional ones.The system is based on the technology of real-time simultaneous amplification and testing (SAT),with which the time of amplification reaction can be greatly reduced.Since the technology of simultaneous amplification and testing still cannot avoid the false negative results,a dual-channel optical system is designed based on the technology to offer accurate detection of the characteristics of sample.Real-time monitoring of negative ones is also achieved.Experiments demonstrate that the system can indicate false negative results effectively within 30 minutes with high accuracy and good stability.
