固定化乳糖酶的研究

摘    要:用明胶作为固定化乳糖酶的载体,研究比较载体的用量、交联剂的浓度,用酶量、制备的pH值。结果表明,选择150g/L质量浓度的明胶为载体,体积分数为0.5%的戊二醛为交联剂,用酶量为0.5g/L,在pH值为7.2.混和搅拌时间为3min的条件下制备固定化果胶酶.其酶活力回收率可达78.12%,重复回收使用7次后,酶活力还可以保留75%以上,是乳糖酶固定化的一种较好的方法。

Abstract:Gelatin was used as carrier for immobilization of lactase.A comparison was made among the preparing conditions,i.e.the dosage of carrier,the concentration of cross linking agent,the dosage of lactase,the time of mixing and pH.The results showed that the recovery ratio of lactase activity was up to 78.12% when the immobilized lactase was prepared with 150 g/L,gelatin as carrier,mixing 3 minutes,5%(V/V) glutaric acid as cross linking agent and 0.5g/L lactase at pH 7.2.and the enzyme activity retained more 75% after recovering 10 times repeatedly.This suggests that gelatinization is a better method for immobilization of lactase.
