阿尔泰锦鸡儿异黄酮提取工艺及不同部位异黄酮含量测定

摘    要:目的 建立锦鸡儿花部异黄酮的最佳提取工艺, 并测定根、茎、叶、果实中异黄酮的含量。方法 在单因素考察的基础上, 采用响应面试验对锦鸡儿花的提取工艺进行优化, 利用最佳工艺提取根、茎、叶、果实中的异黄酮, 并对其进行方法学考察。结果 锦鸡儿中异黄酮的最佳提取工艺是料液比为1:20, 超声提取时间为40 min, 乙醇体积分数为50%, 超声功率为200 W, 其不同部位的异黄酮含量为叶2.74%, 果实2.23%, 花2.21%, 根0.63%, 茎0.39%, 加样回收率为99.3%, RSD=1.0%。结论 锦鸡儿花、叶和果实中异黄酮含量较为丰富, 根和茎中的异黄酮含量较低, 药材不同部位中异黄酮含量的变化可为该药材的取材和深层开发提供基本数据。

Abstract:Objective To optimize the extraction process of isoflavones from Caragana flowers, and to determine isoflavones in roots, stems, leaves and fruits. Methods On the basis of single factor investigation, the extraction process of isoflavones from Caragana was optimized by response surface test. Isoflavones from roots, stems, leaves and fruits were extracted by the best process and investigated in methodology. Results The optimum extraction process of isoflavones from Caragana was as follows: The ratio of material to liquid was 1:20, the ultrasonic extraction time was 40 min, the volume fraction of ethanol was 50%, and the ultrasonic power was 200 W. The contents of isoflavones in different parts were leaves 2.74%, fruits 2.23%, flowers 2.21%, roots 0.63%, stems 0.39%. The recovery rate was 99.3%, RSD=1.0%. Conclusion The content of isoflavones in flowers, leaves and fruits of Caragana is rich, but the content of isoflavones in roots and stems is lower. The changes of isoflavones in different parts of the medicinal materials can provide basic data for the collection and deep development of the medicinal materials.
