芜菁中性多糖降血糖作用研究的初步探讨

摘    要:目的 初步探讨芜菁中性多糖对糖尿病大鼠的降血糖作用。方法 选取造模成功的SD大鼠,随机分为6组,分别为空白组、模型组、阳性对照组(0.20 g/kg)以及芜菁中性多糖高剂量组(0.20 g/kg)、中剂量组 (0.10 g/kg)、低剂量组(0.05 g/kg)。连续灌胃4周, 空白组喂养普通饲料, 其余组喂养高脂高糖饲料, 每周测一次体重与空腹血糖, 测定各组大鼠口服葡萄糖0、30、60、120 min后的血糖值, 计算曲线下面积(area under curve, AUC), 测定肝糖原含量。结果 芜菁中性多糖高、中、低剂量可明显改善STZ与高糖饲料所致糖尿病大鼠的糖代谢功能(P<0.05或P<0.01), 显著降低模型大鼠血糖(P<0.05), 多饮多尿症状明显改善, 可显著增加大鼠的糖耐量(oral glucose tolemnce, OGT) (P<0.01)。结论 芜菁中性多糖对糖尿病模型有降血糖作用, 高剂量组降血糖效果略强于中、低剂量组。

Abstract:Objective To investigate the hypoglycemic effect of neutral polysaccharide from Brassica rapa L. on diabetic model rats. Methods SD rats with successful modeling were randomly divided into 6 groups, including blank group, model group, positive control group (0.20 g/kg), high dose group (0.20 g/kg), medium dose group (0.10 g/kg) and low dose group (0.05 g/kg). Gavage was continued for 4 weeks. The blank group was fed with common feed, and the other groups were fed with high-fat and high-sugar feed. Body weight and fasting blood glucose were measured once a week. The blood glucose values of rats in each group after oral glucose 0, 30, 60 and 120 min were measured, the area under curve (AUC) was calculated, and the liver glycogen content was determined. Results High, medium and low doses of neutral polysaccharide from Brassica rapa L. could significantly improve the glucose metabolism of diabetic rats induced by STZ and high-sugar diet (P<0.05 or P<0.01), and significantly reduced blood glucose in model rats (P<0.05). The symptoms of polydipsia and polyuria were significantly improved, and the glucose tolerance (OGT) in rats was significantly increased (P<0.01). Conclusion The neutral polysaccharide from Brassica rapa L. has hypoglycemic effect on diabetic model rats, and the high-dose group was more effective than the middle and low dose groups.
