摘    要:再造烟叶总植物碱测定采用YC/T 468-2013的方法,但是在测定时会遇到缓冲溶液B很难溶解的问题,即使加热超声溶解后,也会出现结晶析出,容易导致管道堵塞。为了解决此问题,通过减少缓冲溶液B中磷酸氢二钠的用量的方法测定再造烟叶总植物碱含量,并将测定结果与标准方法测定结果进行对比分析。结果表明:缓冲溶液B磷酸氢二钠用量为130 g时,对再造烟叶总植物碱测定结果没有影响;改进后的检测方法检出限和定量限分别为0.057 2%和0.202 8%,相对标准偏差为(0.19~0.55)%。改进后的方法适合再造烟叶总植物碱含量的测定。

An Improved Method for Determining Total Alkaloid Content in Reconstituted Tobacco
Abstract:The YC/T 468-2013 method is usually used to determine total alkaloid content in reconstituted tobacco, but it is difficult to dissolve the buffer solution B during determining. Its crystal precipitation can also appear after dissolving by heating and ultrasonic, which will cause the clogging in the pipe. In order to solve the problem, the amount of disodium hydrogen phosphate in the buffer solution B was decreased, and determined results of the total alkaloid content in reconstituted tobacco were compared with analyzed results. The results show that: when the dosage of disodium hydrogen phosphate in buffer solution B is 130 g, it has no effect on determined result of total plant alkaloid content; the detection limit and the quantitative limit of improved detection methods respectively are 0.0572% and 0.202 8%, and relative standard deviation is 0.19%~0.55%.The improved method is suitable for determining the total alkaloid content in reconstituted tobacco.
