低温苯胺降解菌固定化菌丝球方法与特性

摘    要:为了提高低温条件下含苯胺废水的生物处理效率,采用变温驯化法从活性污泥中分离出低温(7℃)苯胺降解菌JH-9,研究了其降解特性及生物质载体固定化方法.结果表明,JH-9属于乙酸钙不动杆菌属,能以苯胺为唯一碳源和氮源。可耐受苯胺质量浓度1 000 mg/L以上,低温(7℃)下,培养52 h对初始质量浓度为150 mg/L的苯胺去除率可达100%.以黑曲霉Y3作为生物质载体,采用同时接种霉菌和JH-9细菌的培养固定化方法比先后接种霉菌和JH-9细菌的培养固定化方法获得混合菌丝球的培养时间短,且在相同时间内可固定的细菌量大.混合菌丝球的直径、质量和体积均比空白霉菌菌丝球的大.混合菌丝球对含苯胺废水表现出了较好的降解效能,在15℃下低温培养66 h即可将初始质量浓度为150 mg/L的苯胺完全降解,固定化后的功能菌(JH-9细菌)仍然保持了原有的活性.由此,本研究提出了同时培养法.此方法将会有效解决传统载体传质效率低、固定生物量低、成本高等问题,具有广阔的应用前景和工程推广价值.

Characteristic and Immobilized Mycelial Pellets Method of Aniline-degradation Bacteria at Low Temperature

Abstract:In order to enhance the biodegradation efficiency of aniline wastewater at low temperature,we separated low-temperature aniline biodegraded bacteria JH-9 from wastewater treatment factory activated sludge with changing temperature domestication method.Biodegradation character and immobilization for JH- 9 were studied.The result shows that strain JH-9 classified as Acinetobacter-calcoaceticus.It can utilize aniline as the sole source of carbon and can endure 1 000 mg/L of aniline.After 52 h cultured the degradation rate of strain JH-9 can achieve 100% when provided with 150 mg/L aniline with low temperature at 7℃. We use Aspergillus Y3 as carrier.Mixed mycelial pellets forming by simultaneity incubated has less incubative time than which were asynchronously incubated.Besides,the number of immobilized bacteria,size,weigh and volume of mixed mycelial pellet are all bigger than that of mildew mycelail pellet.This is the first study reporting the new immobilization method-simultaneity incubated method.This method can solved the problem of conventional carrier,such as low efficiency mass transfer,less biomass and high cost,and will have great future in terms of application.Mixed mycelial pellets have high aniline biodegrading efficiency,after 66 h cultured the degradation rate of mixed mycelial pellets can achieve 100% when provided with 150 mg/L aniline with low temperature at 15℃.Strain JH-9 can retain activity after immobilization.
