肝片吸虫18.6kD蛋白抗原双向电泳及免疫印迹分析

摘    要:从分子水平上研究了肝片吸虫的抗原组分,用制备性SDS-PAGE纯化ES18.6kD蛋白抗原,并用它成功制备了单因子兔抗血清,用这种抗血清来分析肝片吸虫各部分抗原,证实各部分抗原的18.6kD多肽具有相同的抗原决定簇,而且这种抗原决策簇不存在于其它分子量的多肽中,体抗原(BA)和分泌排泄抗原(ES)的双向电泳(Two-dimention Electrophoresis,2-D)分析研究表明,肝片吸虫同一种分子量的蛋白质大多由多种等电点不同的多肽组成,同时也显示了体抗原和分泌一排泄抗原分子量相同的蛋白质所包含的多肽数量和性质是有差异的。

Abstract:Extracts of F. hepatica-excretory secretory products(ES), and body extracts (BA) of adult worm were analysed by protein electrophoresis (Two-dimention electrophoresis),and the antigen components of Fasciola hepatica was analysed upon a molecular level.The paper reported one monospecific antiserum(rabbit) prepared with 18.6kD peptide of ES purified by preparing SDS-PAGE, and its immunoblotting with four parts of extracts. The results showed 18.6kD in four parts of extracts had the same antigen determinant, while other peptides in different M.W. didn't.Two-dimention electrophoresis (2-D) of ES and BA indicated that many peptides in different PI had same M.W.,and differences were observed between ES and BA.
