洗脱剂极性对硅胶柱分离人参皂苷C-K的影响

摘    要:酶转化后的人参皂苷C-K含有杂质及其他少量皂苷,因此要得到纯度较高的人参皂苷C-K,要对其进行除杂与分离。以酶转化人参皂苷C-K为原料,采用石油醚除杂,除杂后利用硅胶柱层析进行分离得到人参皂苷C-K,并研究了洗脱剂极性对硅胶柱分离人参皂苷C-K的影响。石油醚除杂率为15%。采用3种不同极性洗脱剂分别对30 g人参皂苷C-K进行分离比较,发现V(氯仿)∶V(甲醇)=9.5∶0.5作为洗脱剂时,得到人参皂苷C-K 11.25 g,得率为37.5%,分离效果较好;V(氯仿)∶V(甲醇)=9.0∶1.0作为洗脱剂时,仅得到人参皂苷C-K 3.02 g,得率为10.07%,分离效果较差;采用V(氯仿)∶V(甲醇)=(9.5∶0.5)+[V(氯仿)∶V(甲醇)=(9.0∶1.0)]联合作为洗脱剂时,得到人参皂苷C-K 6.40 g,得率为21.33%,分离效果一般。V(氯仿)∶V(甲醇)=9.5∶0.5洗脱得到的产物经高效液相色谱检测,发现稀有人参皂苷C-K的纯度可达98%以上。

Abstract:The ginsenoside C-K obtaining from enzymatic conversion contains a small amount of impurities as well as other saponins.To obtain the high purity ginsenoside C-K,petroleum ether extraction and silica gel column chromatography were used and the polarity effects of the eluant on silica gel column separation of ginsenoside C-K were studied.Three kind of eluant with different polarity were used to separate 30 g ginsenoside C-K.When chloroform∶methanol is 9.5∶0.5 the yield was 37.5%,chloroform∶ methanol is 9.0∶1.0 the yield was 10.07%,and chloroform∶methanol is 9.5∶0.5 plus 9.0∶1.0 the yield was 21.33%.When the ginsenosides C-K was eluted with chloroform∶methanol=9.5∶0.5 on HPLC the purity was about 98%.
