黑曲霉电解法制备甲壳素的研究

摘    要:本试验以实验室发酵的黑曲霉菌丝体为原料,采用电解法从中提取甲壳素,再将甲壳素碱法脱乙酰转化为壳聚糖。通过单因素试验和正交试验分析了电解时间、电压、碱浓度及料液比对壳聚糖的脱乙酰度、得率、粘均分子量等指标的影响,以此间接反映电解法各因素对甲壳素产品的影响。黑曲霉电解法制备甲壳素的最优条件为:电解时间1.25h,NaOH浓度3%,电压8V,液料比12:1。试验证明,利用电解法从黑曲霉中提取甲壳素实践上是可行的。

Abstract:Chitin was prepared by electrolysis from mycelia through small-scale fermentation of Aspergillus niger in this paper, while the obtained chitin was further deacetylated into chitosan by alkaline. Through single-factor experiments and orthogonal experiment, the effects of some factors on the degree of deacetylation, yield and molecular weight of the chitosan product were studied. The optimized conditions for preparing chitin product from A. niger mycelia by electrolysis were as follows: time of electrolysis 1.25h, concentration of NaOH 3%, voltage 8V and ratio of solvent to meal 12:1. It is found that preparing chitin from the A. niger by electrolysis is practically feasible.
