四环素人工抗原的制备与鉴定

摘    要:根据四环素的化学结构特点,四环素与对氨基苯甲酸通过重氮法合成半抗原.经过酸化,乙酸乙酯萃取提纯,半抗原通过质谱鉴定.通过活化酯法,半抗原分别与BSA和OVA连接制备人工免疫原和包被原,抗血清的效价分别为1:70000和1:32000,为制备四环素人工抗原提供一种更为有效的途径,进而建立酶联免疫快速检测方法,为最终实现四环素检测试剂盒的研发奠定基础.

Abstract:A new synthesis way of hapten for tetracycline base on the formation of its diazo derivatives with acidification, extraction by ethyl acetate. Then,happen TC was covalently attached through its carboxylic acid moiety to the carrier protein (BSA,OVA) by means of the active ester method for preparing immungens and coating antigen,the titer of serum were 1 : 70 000 and 1 : 32 000,respectively. The purpose of this artificial is to provide an effective way to preparate haptens for tetracycline. This study laid a well foundation for the control for the control of tetracycline by ELISA method.
