大豆异黄酮纯化工艺研究

摘    要:以大豆为原料,乙醇为溶剂,提取大豆异黄酮,探讨大豆异黄酮提取物纯化工艺,以寻求大豆异黄酮纯化最佳工艺方法。先将大豆异黄酮粗提物进行脱脂、脱蛋白初步处理,再结合大孔树脂吸附与乙酸乙酯萃取精制,最后冷冻结晶得到产品。结果表明:大孔树脂吸附与解吸最佳工艺条件为:上样液浓度0.15 mg/mL,上样pH值为4.5~5.5,上样量为4.5BV,吸附流速为1.5ml/min,静态吸附5h,解吸剂80%乙醇,解吸流速0.5ml/min,并以三波长紫外分光光度法分析检测,得到大豆异黄酮产品纯度可达82.5%。

Abstract:Based soybean as raw materials, ethanol as the solvent, the isoflavone was extracted from soybean flours, and the best purification process was discussed. After the initial treatment of removing fat and proteins in the soybeans, using the method of solvent extraction, resin absorption to purify, fi- nally the products were gained from frozen and crystal. Results show:the best conditions of adsorption and desorption were, the concentration of sample 0.15 mg/mL, pH 4.5~5.5, dosage 4.5 BV, adsorp- tion flow rate of 1.5ml/min, time of the static absorption 5 h, desorption solvent 80% ethanol, desorp- tion flow rate of 0.5ml/min. The purity of soybean isoflavones products can reach 82.5% by UV scan- ning.
