鱼油对母鼠及新生大鼠海马组织NR1、c-fos、CREB基因表达的影响

摘    要:研究鱼油对母鼠与新生大鼠海马组织中NR1、CREB、c-fos基因表达的影响。将64只成年SD大鼠随机分成阴性对照组和鱼油低剂量150 mg/(kg·d)]、中剂量组300mg/(kg·d)]和高剂量组600 mg/(kg·d)],连续饲养60 d,使用实时定量PCR技术检测母鼠以及新生大鼠海马组织N-甲基-D-天门冬氨酸受体亚基(NR1)、即刻早期基因(c-fos)、cAMP反应元件结合蛋白(CREB)mRNA的表达。结果表明,与阴性对照组相比,鱼油低剂量组和中剂量组的母鼠与新生大鼠海马组织中NR1、CREB、c-fos基因的mRNA表达量显著提升,鱼油高剂量组母鼠与新生大鼠海马组织中NR1、CREB、c-fos基因的mRNA的表达量没有明显变化。孕期和哺乳期的饮食中添加一定剂量的鱼油,能够提高母鼠与新生大鼠海马组织中NR1、CREB、c-fos基因的表达。

Effect offish oil on expression of NR1, c-fos and CREB in hippocampus of female rats and newborn rats

Abstract:To investigate the effect of fish oil on mRNA expression of N-methyl-D-aspartate receptor subunit (NR1), immediate early gene c-fos and cAMP responsive element binding protein(CREB) in hippocampus of female rats and newborn rats, 64 adult SD rats were randomly divided in- to negative control group, low dosage fish oit group 150 mg/(kg·d)], mid dosage fish oil group 300 mg/(kg·d)] and high dosage fish oil group 600 mg/(kg·d)]. After continuously feeding for 60 d, Real-time quantitative PCR was used to test the mRNA expression ofNR1, c-fos and CREB in hippocampus of female rats and newborn rats. The results showed that compared with the negative control group, the mRNA expression of c-fos, CREB and NR1 in the hippocampus female rats and newborn rats with low dosage and mid dosage offish oil group increased obviously. But there was no significant difference in the mRNA expression of c-los, CREB and NR1 in the hippocampus female rats and newborn rats with high dosage fish oil group. The optimal dosage of fish oil supplementation during the prenatal and suckling period could increase the mRNA expression of c-fos, CREB and NR1 in hippocarnpus of female rats and newborn rats.
