双酶协联糖化纤维素的条件优化

摘    要:本文以双酶协联糖化技术处理粉渣中纤维素,并利用单因素和正交试验考察菌种混合比例、接种量,协联糖化温度与协联糖化时间对纤维素糖化率的影响,结果表明,双霉协联糖化的最优条件为:少孢根霉与里氏木霉混合比值为1.5,接种量为1.8×108个孢子/ml,协联糖化温度为34℃,协联糖化时间为72h,在此工艺条件下纤维素糖化率高达82.3%。纤维素酶活力达到645.5U/ml~664U/ml,与传统单菌种发酵的糖化技术相比,提高了160U/ml~280U/ml。

The Research of Optimization Conditions on Saccharification of Cellulose Coupling with Trickoderma reesei and Rkizopus oligosporus

Abstract:The optimization conditions on saccharification of cellulose coupling with Rhizopu- soligosporus and Trichoderma reesei were researched. Using the method of single factor and orthogonal, mix- ing ratio of strains, inoculum, saccharification temperature and saccharification time were determined on the effect of saecharification rate of cellulose. The optimums aeeharifieation conditions were showed that the mixing ratio of Rhizopus oligosporus and Trichoderma reesei 1.5, the inoculum 1.8Хl08spores/ml, the sacchar- ification temperature 34℃,the saeeharification time 72h. In this condition, the rate of saccharification of cel- lulose was 82.3%.Cellulase activity was 645.SU/ml-664U/ml, compared with the traditional biology sacchar- ification with single strain, cellulase activity improved 160U/ml-280U/ml
