酿酒酵母摇瓶发酵生产麦角固醇的条件优化及提取方法研究

摘    要:通过多因素条件优化实验确定的酿酒酵母摇瓶培养生产麦角固醇的最佳发酵条件是:28℃,pH 6.0,装液量为45 mL(250 mL),接种量8%,发酵48 h,麦角固醇的含量达到1.36%,产量高达19.95 mg/100 mL。采用乙酸酐显色法进行麦角固醇萃取剂的选择及萃取条件的确定。实验表明,丙酮为优选萃取剂,最佳浸提固液比(菌体质量与丙酮体积比)为1∶25。

Studies on fermentation conditions of ergosterol and its extracting methods
Abstract:The fermentation conditions of ergosterol and its extracting methods were studied in this article. Through orthogonal experiments, the best fermentation conditions were obtained:28℃ ,pH 6. 0,45 mL volume of liquid,8% innoculum,and 48 h fermentation time,the ergosterol content reached the maximum 1.36% ,and the yield of ergosterol reached the maximum 19. 95 mg/100 mL. In the extracting experiments , we employed acetic anhydride test as a fast and direct method to screen the extracting conditions, the result showed that acetone was the best, and the liquid-solid ratio was 1:25.
