酶联免疫法检测猪肉中呋喃唑酮代谢物残留

摘    要:通过检测猪肉中呋喃唑酮代谢物的残留以评价试剂盒的性能,采用酶联免疫法对猪肉中的呋喃唑酮代谢物残留量进行测定。结果表明:该方法的线性检测范围为0.025~2.025μg/L,最低检测限为0.1μg/kg,样本添加回收率为78.2%~100.3%,变异系数为4.7%~10.3%,与呋喃唑酮的交叉反应率为16.3%,与其他药物的交叉反应率均小于1%,且对实际样本的检测结果与液相色谱-串联质谱法基本一致。该法灵敏准确、重复性好、特异性高,适用于猪肉中呋喃唑酮代谢物残留的检测。

Abstract:An enzyme-linked immunosorbent assay (ELISA) was proposed to determine residual furazolidone metabolite in pork using a commercially available ELISA kit. The results showed that the sensitivity and detection limit of the assay were 0.025-- 2.025 μg/L and 0.1 μg/kg, respectively. The recoveries of standard additions of furazolidone metabolite averaged 78.2% -- 100.3% across three levels (n = 3), and the coefficients of variation were 4.7%--10.3%. The cross-reactivity towards furazolidone was 16.3%; as for other drags investigated, the cross-reactivity was below 1%. Analytical results obtained using the assay were in substantial accordance with those from liquid chromatographic tandem mass spectrometry (LC-MS/MS) determination. The assay was sensitive, accurate, repeatable, specific and suitable for the determination of furazolidone metabolite residue in pork.
