玉米高F值寡肽的分离纯化及抗氧化活性研究

摘    要:目的研究玉米高F值寡肽混合物的分离纯化及抗氧化作用。方法采用碱性蛋白酶Protex 6L、中性蛋白酶Protex 7L和木瓜蛋白酶3种酶复合酶解玉米黄粉(CGM),经超滤得到寡肽混合物,用Sephadex G-15凝胶分离纯化,测定寡肽混合物及其分离纯化组分的相对分子质量(Mr)及抗氧化活性。结果寡肽混合物的F值为28.40。寡肽混合物经Sephadex G-15测定其Mr为231~2392,并分离得到3个组分:P1、P2和P3,其Mr分别为2392,1581,231。寡肽混合物对邻苯三酚自氧化的抑制率达73.31%,3个组分对邻苯三酚自氧化的抑制率分别为22.10%,18.86%和75.75%,抗氧化活性最高的为二肽组分P3。结论玉米高F值寡肽混合物有良好的抗氧化活性,P3为主要抗氧化组分。

Study on Separation Technology and Antioxidant Activity of Corn High F Value Oligopeptide

Abstract:Objective To research the separation and purification technology of corn high F value oligopeptide and its antioxidant effect. Methods Combination of three hydrolases(Protex 6L, Protex 7L and papain) were used to hydrolyze corn gluten meal(CGM), oligopeptide mixture were acquired by ultrafiltration, then was separated and purified by Sephadex G-15. The relative molecular weight(Mr) and antioxidant activity of the oligopeptide mixture and separated fractions were determined. Results The F value of the oligopeptide mixture was 28.40. The oligopeptide mixture was separated into three fractions, including P1, P2 and P3. The Mr of the mixture was 231-2392, and the Mr of P1, P2 and P3 were 2392, 1581 and 231, respectively. The inhibition rate of the oligopeptide mixture on autoxidation of pyrogallol was up to 73.31 %, and the autoxidation inhibition rates of P1, P2 and P3 were 22.10 %, 18.86 % and 75.75 %, respectively. P3, the bipeptide fraction, was of the highest antioxidant activity. Conclusion The high F value oligopeptide mixture possesses outstanding antioxidant activity, and P3 is the main antioxidant component.
