基于静电纺丝法构建用于药物可控释放的醋酸羟丙甲基纤维素琥珀酸酯纳米材料的研究

摘    要:醋酸羟丙甲基纤维素琥珀酸酯(HPMCAS)作为一种典型的肠溶材料在药物可控释放领域具有极大潜力。采用静电纺丝法制备HPMCAS微纳米纤维并对其纺丝参数进行优化。通过研究溶液浓度、纺丝电压、纺丝距离及混合溶剂配比的影响行为,确定制备HPMCAS纳米纤维的最佳工艺参数。通过正交实验及单因素变量设计获得最佳实验条件为:溶液浓度12wt%,电场强度20kV,纺丝距离20cm,溶剂比例CH3OH∶DCM=4∶6。该条件下获得的纤维形貌良好、分布均匀,平均直径为363nm。将为HPMCAS纳米纤维作为智能pH控制药物缓释/控释材料提供理论和实验基础。

Abstract:As a typical pH sensitive polymer with biocompatibility, hydroxypropyl methylcellulose acetate succinate (HPMCAS) is a promising material for application in controlled drug release. In this paper, HPMCAS micro-nanofibers were prepared through electrospinning. The optimum condition was obtained by researching the effective of the parameters such as solution concentration, applied voltage, distance and ratio of mixed solvent on fiber morphology and diameter. Through an orthogonal and a single factor experiment, the optimum parameters were determined as a polymer concentration of 12 wt %, a voltage of 20 kV, a collector distance of 20 cm and a solvent ratio (CH3OH/DCM) of 4/6. As a result, the average diameter of fibers is 363m and with good uniformity and smooth morphology. The results will provide theoretical and experimental basis for using HPMCAS nanofibers as intelligent ph-controlled drug sustained-release/controlled-release materials.
