肝素钠精制工艺研究

摘    要:采用过氧化氢二次氧化法精制肝素。在该过程中采用低温离心技术,解决在除酸性蛋白过程中,沉淀不易滤除的困难。并通过实验确立酶解结合氧化法精制肝素新工艺,克服粗品肝素钠中残留杂蛋白在纯化过程中很难被除去的缺点。在粗品肝素钠中加入胰蛋白酶对这些杂蛋白进行酶解,再结合氧化除杂过程,从而提高精品肝素钠的效价和效价回收率。

Abstract:The purification of heparin by double oxidation process with H 2O 2 was investigated. The centrifugal technique under low temperature was used to solve the difficulty of removing of the precipitation. The the crude heparin sodium salt was degraded by adding trypsin, so that the residual proteins were removed from heparin. The results showed that the improved process, which combined oxidation and degradation, can raise the potency and recovery of heparin sodium salt.
