Determination of Mitiglinide in Human Plasma by LC-MS/MS

MitiglinideinhumanplasmawasdeterminedbyLC-MS/MS.Mitiglinideandtheinternalstandardwereextractedfromplasmabyacetonitrile,whichwasusedasdeproteinatedsolvent,andthenseparatedonaKromasil-C₁₈ column(50mm??4.6mm??5??m).Themobilephaseconsistedofacetonitrile??1mmol•L^-1 ammoniumformate(containing0.1%formicacid)maintainedat30??.Theflowratewas0.6mL•min^-1,and20??LaliquotofresidueswereinjectedintotheLC-MS/MSsystem.Detectionwascarriedoutbymultiplereactionmonitoringonan3200QtrapLC-MS/MSsystem.Theassayislinearovertherange3.0-3000.0??g•L^-1 withalowerlimitofquantitationof3.0??g•L^-1.Intra-dayandinter-dayprecisionarebothlessthan15%,respectively.Therelativedeviationisintherangeof-1.40%-2.07%.TherecoveriesofMitiglinideare(98.2??7.6)%,andstabilitiesaregood.Itisarapid,sensitive,selectiveandreliablemethodforthedeterminationofMitiglinideinhumanplasma.TheassaycanbeappliedforthedeterminationofMitiglinideinhumanplasmaandthestudyonpharmacokinetics.
