摘    要:为探索常压室温等离子体(ARTP)诱变对不同基因型甘蔗胚性愈伤组织的合适诱变时间和筛选草甘膦浓度组合,以胚性愈伤组织活愈率为指标,研究不同诱变时间(80~180 s)和不同草甘膦浓度(0.5~2.5μmol/L)对含有大茎野生种血缘后代(粤糖94-128、ROC22)、斑茅血缘后代(崖城07-71)和割手密血缘后代(粤糖93-159、粤糖00-236)这3种血缘甘蔗胚性愈伤组织的影响情况。结果表明:不同时间诱变处理下甘蔗胚性愈伤组织平均活愈率差异显著,取含割手密血缘的后代材料和含大茎野生种血缘的后代材料来诱变的突变率高;为筛选到高抗材料,合适诱变时间为120 s,草甘膦最高筛选浓度为2.5μmol/L。研究结果为以后进行高抗草甘膦诱变提供了试验依据。

Abstract:In order to explore the appropriate mutagenesis time and the glyphosate concentration of sugarcane embryogenic callus by atmospheric room temperature plasma(ARTP) mutagenesis, the test was conducted to get mutants with high resistance to glyphosate. The survival rate of embryogenic callus was used as the index to investigate the effects of mutagenesis time(80~180 s) and glyphosate concentration(0.5~2.5 μmol/L) on the sugarcane embryogenic callus of three bloods with S.robustum Brandes and Jesw. ex GrassⅠ(Yuetang94-128, ROC22), E.arundinaceum(Retz.) Jeswit(Yacheng07-71) and S.spontaneum L.(Yuetang93-159, Yuetang00-236). It was found that the average survival rate of sugarcane embryogenic callus was significantly different at different mutagenesis times. The higher mutation rate was observed for the consanguineous relatives of S.spontaneum L. and S. robustum Brandes and Jesw. ex GrassⅠ. To obtain the mutants with high resistance to glyphosate, the appropriate mutagenesis time was 120 s and the maximum concentration of glyphosate was 2.5 μmol/L. The results would provide the experimental basis for further study to get mutants with high resistance to glyphosate.
