正交优化燕麦β-葡聚糖提取及其分子特性研究

摘    要:为确定提取燕麦中β-葡聚糖的最佳工艺条件,以分光光度法测定得到的燕麦β-葡聚糖含量为指标,采用单因素实验和正交试验设计,分别考察pH值、液料比、温度和提取时间对提取率的影响.由此知提取燕麦中的β-葡聚糖的较佳工艺是,液料比为25,pH值为11时80℃水浴提取4h.用凝胶色谱测定纯化后β-葡聚糖平均分子量,得其平均分子量为9.697×10^5D.

Abstract:The extraction condition for β-glucans from oat bran was optimized. UV-spectrophotometry was used for the determination of β-glucans. The effects of β-glucans extraction were analyzed in several single factor experiments and orthogonal experiment. The results showed that the optimum conditions as follows: extraction temperature is 80℃, ratio of liquid to material is 25, pH is 11, and extract time is 4 h. The molecular weight of β-glucans after purified was obtained using high performance gel permeation chromatography (GPC), and the average molecular weight is 9. 697×10^5D.
