生物表面活性剂发酵条件的响应面分析优化

摘    要:本文利用Plackett-Burman设计法和响应面分析法,对产生物表面活性剂的采油菌株DN4-3的发酵条件进行优化,筛选出三个显著影响因子(葡萄糖用量、酵母浸粉用量和氯化钠用量)。在此基础上,通过最陡爬坡实验和响应面分析,研究了显著因素的最佳水平及其交互作用,确定了最佳发酵条件。最佳发酵条件为:葡萄糖27.83 g/L、酵母浸粉2.73 g/L、NaCl 2.37 g/L、KH2PO4 9 g/L、K2HPO4 3 g/L、CaCl2 0.16 g/L、MgSO4.7H2O 0.5 g/L、接种量4%、种龄16 h、装液量250 mL/500 mL、温度37℃、初始pH值7.0、摇床转速160 r/min。采用优化后条件进行摇瓶发酵,产表面活性剂1025.98 mg/L,与预测值很接近,比优化前提高了35.43%。
